Tauopathy pathogenesis

Our research focuses on probing the pathogenesis of tauopathy. Tauopathy is a category of diseases of which the pathogenesis processes are all related to abnormal tau protein aggregation, which includes Alzheimer’s diseases and various parkinsonisms. Tau protein normally associates with microtubules to stabilize their structure, but it could undergo complex posttranslational modifications, causing its detachment from microtubules and forming aggregates under pathological conditions. Using Drosophila to expressing different human tau isoforms mimicking different posttranslational modifications, I could investigate which modifications are critical for modulating the protein’s toxicity and provide insights to some unsolved questions related to the posttranslational modifications of Tau. This study may help to pave the road for therapeutic development.

Tau蛋白致病機轉研究

我們的研究主要聚焦在tauopathy的病理機制。Tauopathy是一個疾病類型的泛稱,其發病機制都與一種名為tau的蛋白異常聚集有關,而此類疾病包含阿兹海默症以及不同的類帕金森氏症。正常情况下,tau蛋白會與微管蛋白結合並穏定其結構,然而在病理狀態下,它會經過不同的蛋白後修飾作用使得它從微管上脱落並變得傾向形成聚集,而導致神經功能失調。我利用果蠅表達不同亞型的人類tau蛋白以模仿不同的後修飾作用,來探究哪些蛋白修飾會對tau的細胞毒性有顯著影響。這項研究可以找到對tau蛋白後修飾所產生的病理機制的答案,並提供tauopathy這類疾病未來藥物開發的基礎。
